Dear all,
Can anyone recommend well-tolerated solvents for short infusion (10
min) into dogs or point me in the right direction of where I might
find this information?

DMSO, pharmasolv are some of the solvent that help to dissolve the
difficult-to-dissolve molecules, as well as are safe to certain
concentration. You need to evaluate the concentration that may lead to
toxic effects that may interfere with toxic profile of the molecule.

Dear Fredrik,
Poly Ethylene Glycol 300 can be used for IV
applications.This is a safe excipient.However if the
drug is not soluble in PEG then primary solubilisers
like Pharmasolve or 2- Pyrole maybe used in quantities
upto 5%v/v of the formulation further diluting the
same with PEG.But an aqueous component upto 25-30% is
a minimum requirement in the formulation.

Dear Frederik ,
You can use DMSO , pharmasolve , co-solvent approaches and go
through chemistry part of the molecule for selecting appropriate
excipient for enhance the solubility without depending on large
extent on only solvents in the formulation .Run the experiment with
placebo formulation for differential toxic & safety effect of
infusion vehicle and drug .Refer IIG ( Inactive ingrdients guide ) .

Dear Dr. Frederik,
DMSO in high quantities precipitates Neurotoxicity. There are a lot
of other inactives whose toxic limit is given in the www.fda.gov.cder
site. Trying with liquid PEGs up to 22%w/v is acceptable. With DMSO
the limit is narrow approx 3 - 5 %v/v. Preferably perform a placebo
study to assess the toxicity when employing DMSO.

Dear Frederik and Korlapati,
When planning to use DMSO or other potentially toxic co-solvents, it
is good to think all the way ahead to dosing humans. If no
alternatives come to mind, then this may influence your plans. If
one, such as Cremophore, is possible, then you may wish to
incorporate this into your current plans, (to avoid duplicating some
of your work in future "bridging studies").
