Dear all!
I am now confused with one problem. Do PPK could be used to predict
the tissue concentration in the target population? I found the NONMEM
method is designed for calculating the kinetics of drug in blood, I
don't know whether if it could be used in predicting tissue kinetics.

My questions (related to PharmPK inquiry of Nov. 20 by:

(1) TARGET tissue identification and target pharmacokinetics, are
these guideline-driven parameters? Is in vivo target identification
required for the approval of a new drug?
Such general requirement, as far as I know, does not exist. ADME
studies require information on tissue DISTRIBUTION. However, this
does not specify whether the distribution is related to target,
transport, metabolism, excretion - all so-called sites of loss, or
sites of action.
(2) Can drug target tissue concentrations be assessed from plasma
(blood) concentrations? What is the assurance (evidence) that such
practice would be realistic and not misleading?
Any clarifying comment is appreciated. There seems to be some confusion.
