Drug use is a problem in the acute porphyrias because many drugs can
precipitate attacks. This may, at least in part, be a concequence of a
reduction of the free hepatic heme pool after induction or mechanism based
inhibition of CYP450. Even CYP450 effects which wil not lead to clinically
meaningfull drug interactions may be of importance to porphyria patients.
What are the most comprehensive information sources for CYP450 induction or
(especially) mechanism based inhibition? Any good reviews? I am also
interested in structural chemical features associated with mechanism based
inhibition.
