Colleagues,
We are looking into incorporating a 'high-throughput'
in vitro permeability/absorption assay into our
existing battery of in vitro DMPK tests. Can someone
provide insight as to the relative merits/drawbacks of
Caco-2 vs. MDCK cells? In addition to the ability of
each to predict human absorption, factors such as
cost(!), P-gp/3A4 levels, simplicity of the assays
(3-day vs. 21 day) and general acceptance of each
assay in the DMPK community will be important in our
decision. Any insight, advice or specific references
that you may have on the topic will be greatly
appreciated.
