Hello all:
I am interested in examples of approved drugs characterized by low
clearance. The characteristics should include high lipophillicity, large
distribution volume, low or slow metabolic clearance and long terminal
half-life. Some examples would include amiodarone or warfarin. It would be
very helpful if a reference for the pharmacokinetic properties was included.
My real interest is in the issue of accumulation and its impact on clinical
trial design. I appreciate any information offered.

- Vitamin D3 (cholecalciferol) - t1/2,z=1 week (one main metabolite)
- 25-hydroxycholecalciferol(calcidiol) - t1/2,z=3 weeks (2 main metabolite)
