sub:Instability of benzofuraxone ring in blood

I come a cross this type of problem while doing pharmacokinetic
studies of compounds having benzofuraxone ring.During analysis of
Intra Venous dose samples,even intial 1 min time point sample did
not show presence of parent compound. I found nitro,amino derivative
of parent compound instead of parent benzofuraxone ring
structure.During extraction efficieny,spiking in rat plasma,followed
by organic solvent extraction,recovery was more than 90% and
reproducible.

This experiments followed by in-vitro stability in rat blood.We found
similar results as we got in pk studies.
What could be the possible reasons?
