Hi Jie,
When the drug is given by i.p. route it by passes the gut and will be
subject to hepatic 1st-pass metabolism.

Hi Kassem and others,
Thanks! What's the percentage of absorbed drug after I.P. administration
subject to hepatic 1st-pass metabolism? With respect to this issue, I
have
asked more than 10 senior scientists inside and outside our company
whose
majors are PK. The answer varies from 0 to 100%. From my anatomy
knowledge,
at least part of the drug is absorbed though the veins inside the wall
of
abdominal cavity therefore bypass the 1st-pass metabolism. I wouldn't
accept
the answer of 100%. The answer to my question will tell us the exact
impact
of 1st-pass metabolism on I.P dose, especially when we compare I.P.
versus
Portal vein injection in case the compound is completely (~100%)
absorbed
from the I.P. injection site.
