Dear Shailaja
We worked on the pharmaokinetics of i.m nemesulide in animals. Just go
through these publications.

As far as HPLC method is concern you can try using 0.05M
Ammoniumdihydrogenphosphate and acetonitrile 50:50 using C18 column
with UV detection. In this mobile phase you can estimate
metabolite(hydroxy nimesulide) too. For sample preparation go for SPE
extraction or direct deproteination using 50% ACN, centrifuge and
inject 50µl for quanitification.

Dear Shailaja,
even if some injectable preparation of Nimesulide were proposed in the
past (see Gupta works), the low water solubility of the molecule does
not allows to achieve an acceptable formulation (almost for humans).
The relative bioavailability of Nimesulide is really high and the
first pass effect does not affect the plasmatic concentration.
The half life is not modified by the route of administration.
