Hi all,
Does anyone knows the S-mephenytoin pKa? I'am trying to develop a HPLC
method for to detect and quantify it.

Hi Damaris:
I am not sure what the pka is, but one can use IPA: H20, 0:100, ramp
it up to 60: 40 in 8 min, hold for 3 min, with flow rate of 0.8 mL/min,
.aaa. 225 nm and 40C. An alternative method is to use ACN: H20 or MeOH: H20
as mentioned in the Gentest catalog. Hope this will be useful. Happy

Dear Dr Diaz,
The pKa of phenytoin is 8.3. I don't expect the pka of
S-mephenytoin to differ much, though.
As a rule of thumb, you can run the HPLC using a
mobile phase pH which is of +/-2 units from its pKa
value.
