We have been using the classical colorimetric method to quantify plasma
bilirubin during the course of maximum tolerated dose studies in the
rat and in the dog. Both direct and total bilirubin were assayed with
the "Sigma Diagnostic Kit", where bilirubin reacts with the sulfanilic
acid (the use of a surfactant or not provides the specificity for total
and direct bilirubin, respectively). Analytical results clearly
demonstrate an hyperbilirubinemia (both are increased) without any
histopathological signs. We suspect an analytical interference with our
compound (or metabolites), and this is currently under investigation in
the lab. Actually, the list of drugs that can increase bilirubin
measurement is important an include anabolic steroids, diuretics, MAO
inhibitors, oral contraceptives, salicylates ....... Did you have any
experiences and/or good references on (i) how to manage analytical
interferences using this Kit, and (ii) analytical alternatives (mass
spec ... or others ...) .
