Hi,
I am looking for sensitive assays (and calculations) allowing to determine
estradiol (free fraction, total fraction and SHBG-bound fraction) in biological
matrices.
On a technical and also regulatory point of view, what would be the methods of
choice (immunoassay vs LC/MS/MS or GC/MS) considering that, in our case,
estradiol is the drug product and not only a biomarker ?

I would recommend you to have a look at DELFIA products which have been
successfully used for your application by some pharma companies. DELFIA
method is a sensitive fluoroimmunoassay that has a wide dynamic measurement
range beneficial for this type of assays.
