Subject: Maximum dosage of PEG 400

Hello every one
Could some one tell me, what is the maximum percentage of PEG 400 can be given
to the rats and mouse for PK studies ? Our drug is soluble in PEG 400 100 % but
precipitates when we dilute the PEG to 50 %. It is dissolving in 75 % PEG 400. I
guess, even 75 % of PEG is toxic to the animals.

according to OHS material safety data sheet for PEG400
for the rat
oral LD50 30200 mg/kg
ip LD50 9708 mg/kg
iv LD50 7312 mg/kg
for the mouse
iv LD50 8550 mg/kg
in my experience 25 % PEG400 in saline is tolerated by the mouse. also using PEG200 decreases viscosity for iv administration
