I was playing around with the equations for a simple model of a parent
compound degrading to a first generation metabolite. Assuming simple
first order kinetics for each degradation process, I derived an
analytical expressions for the relationship between the molar formation
fraction (f) of the metabolite and its peak occurrence (p, expressed in
parent equivalents as a fraction of initial mass):
f =p*(hm/hp)^(hp/(hm-hp)), where hp and hm are parent and metabolite
half-lives respectively.
A colleague told me that this relationship is well known but I have not
been able to track down a reference. I am particularly interested to
find out whether such relationships can be extended to second and higher
generation metabolites. Does anyone where I should look in the
literature?
