GastroPlus(tm) is used throughout the industry for this. I believe it
is the only program available that covers controlled release in a
comprehensive way - gastric release, integral tablet, and dispersed
release options, as well as the ability to optimize a release profile
to achieve a target plasma concentration-time profile, all while
accounting for saturable metabolism and transport (if needed),
pH-dependent solubility, regional absorption effects. etc.

Or alternatively, use the generic and flexible approach as used in
NonMem: a
combination of zero- and first-order absorption constants, possibly
divided
over multiple dose compartments.
