Dear all,
There are studies on determination of PCB microsomal metabolites
(PCB-OH) using GC. Why is that these people use PCB-OMe as their
standard and they also derivatize their metabolites to PCB-OMe rather
than using PCB-OH. (even in the papers that they donot use MS as
detector.)

Dear Ahmad,
There are several reasons. First, PCB-OH is an acidic compound which
trails on some chromatographic columns, which broadens the peak shape.
The broader the peak shape, the less resolution one has. Second,
phenolic compounds (like PCB-OH) can oxidize which results in misleading
impurities.
Conversion to PCB-OMe improves chromatographic separations and stops
formation of oxidative by-products. Further, MS ionization improves
substantially with PCB-OMe.
