Dear Frederic,
your statement of "cleaning" the mucosa from the epithelium is very
interesting. In my understanding, the first effect of a hydrophilic and
water soluble surfactant like Tween 80 is on the gel like unstirred
water
layer (e.g. to "wash it away") and not, as most of the literature
states on
the epithelium itself. Once the USW has been loosened, a compound -and
especially a lipophilic one- may more easily penetrate into and through
the epithelium.
Since the mucosa is renewed about every 24 h by the intestinal mucosa
glands, the effect of the excipient on the USW (if that is the only one)
should be fully reversible. Actually, I haven't found too much
literature
regarding this aspect.

Philip,
I agree with your point regarding the reversible effect of the
excipient.
What I mentioned in my previous mail is that a long term treatment
would not
allow the mucosa replacement and the epithelium would be kept in contact
with irritants such as acids. The potential ulceration is not the fact
of
the surfactant but a consequence of its overuse.
