Subject: regarding sample size in preclinical PK

hi all
it is about sampling size during preclinical PK studies in rats. We have performed oral and topical PK studies with n=3 rats/time point for 14points using 2 samples per rat (one cardiac and other terminal from venacava).i want to know how good it is to draw conclusions
from this data. if n=3 is not sufficient what is the minimum samle size for
preclinical studies. any supported reference will be greatly appreciated. for
urine and fecal excretion studies also is there any significance of n=3.
