Hello,
We are developing an LC/MS/MS method for a cytotoxic compound, using
taxol (paclitaxel) as internal standard.
Did anyone experience some stability problems with taxolduring method
validation or sample analysis?
The LC/MS/MS signal of taxol (raw material just dissolvedin the mobile
phase), is surprisingly changing over time (on short intervals : 0 to 5
hours).

It depends in what vessel the samples are made in. The material gets
absorbed into plastic vessels, less so on glass....

Hi, VaNic,
I used Taxol (in 10% methanol) as IS for HPLC analysis and it was quite
stable (for several days), as long as the Taxol solution was stored in
fridge.

Hi Valerie!
Probably your problem depends on the material of vessels used for
samples.
For example standard glassware do adsorb in different amounts
paclitaxel and
also docetaxel. This do not happen with dimethyldichlorosilane-methanol
treated glassware.
Untreated glassware leads to a significant loss of sample and the loss
is
time-dependent.
We measured a drug loss of about 40-50% with not-treated glassware
immediately after preparation of drug solution. This loss significantly
increased already after 30 min.

Paclitaxel solution (prepared in methanol water mixture) is pretty
unstable. I dont have the exact stability data but I remember that
within a day, you see several additional peaks in HPLC-UV in samples
stored at -80C.
