What is the current wisdom when doing a 14C tissue distribution study
in rats regarding perfusing animal to remove "blood" component from
organ or using a correction for residual hemoglobin or neither. Just
compare blood/plasma concentrations to organ concentrations to make
assessments of tissue levels.

Dear Dave,
In my experience, this perfusion method does do too
much plus is messy. It is better to use published
values for blood content of various tissues and do
your correction that way. Is it worth doing this at
all? Depends on the physico-chemical characteristics
of the compound, sites of accumulation and toxicity
profile and target organ for toxicity.
