Hello Everybody,
I am new to this group and before I go to "business" with a question, I
would like to introduce myself.
I am working for a biotech company in Munich (Germany) which works in
the
field of small molecule drug discovery. I am responsible for the
DMPK & Analytics subdepartment.
Currently we are looking into promising compounds which have a
surprisingly
high oral bioavalability. We suspect an active transport (simple
experiments with Caco-2 cells and PAMPA-bilayers confirmed this). Since
we
do not have the necessary inhouse expertise in the transprter field, we
think of approaching skilled contract organizations ("CROs") for this
purpose. Has anybody of you experience with such a CRO,
specialized in the transporter field? We have had already contact with
"standard" CROs like MDS etc. but nobody we have found so far has the
appropriate service on hand.
Thank you very much in advance!

Hallo, Markus
your question is very complex since there are many uptake and efflux
transporters in the gut.
Possible uptake transporters could be PEPT1, ASBT, GLUT 2/3/5, SGLT1,
MCT1,
DTT, LNAAT.
It depends on your compound which one of the uptake transporters could
be
responsible for uptake- and having a certain idea you could specify your
question again.
To my knowledge, there is no CRO covering the complete range of uptake
transporters. You should 1. contact Universities or Institutes with a
certain
proposal for your compound or 2. perform some inhibition experiments in
CACO-2.
