I am trying to incorporate an absorption delay for multiple dose
administrations using rates. I used the SHIFT/ OUTPUT subroutine in a
fortran file as described below. The absorption delay is created by
shifting all doses by the same delay.
Subroutine OUTPUT (Y,T,X)
Implicit None

Include 'model.inc'
Real *8 Y(MaxNOE), T, X(MaxNDE), DelayPar
3. Output equations:
DelayPar = 4
Call SHIFT(P(DelayPar),T)
Y(1) = X(1)/P(2)
This subroutine works well with bolus administrations, however, the
delay does not seem to be applied when rates are used (R(1), R(2),
etc). Although a value is calculated for the DelayPar parameter, this
delay is not applied to the predicted concentrations (visual inspection
of the profiles shows that systemic absorption is beginning at a time
where the delay is predicted to be ongoing). A colleague of mine has
also experienced the same problem.
Has anyone had success with this subroutine using dose administration
by rates? Any comments or suggestions would be appreciated.
