Does anybody have experience administering solids such as gelucire to
rats?

What exactly are you looking for? Yes....capsule administration to rats
can be accomplished (there are some specialized kits out there for
this...the capsule shells are obviously quite smaller compared to most
administered in canines and uses a special delivery device to get past
those nasty rodent teeth). In my previous experience, we have used it
to look at the PK profile of novel antineoplastics after single oral
administration for solid dosage forms, as well as looking at different
solid formulations (i.e. different crystalline batches of material, etc)
to "scout" for a solid form with good dissolution properties before
going into more expensive and limiting canine studies.

Dear Marla,
I have been working on Gelucire 44/14 for enhancement of oral
bioavailability studies.
I have screened these dispersions in rats for oral PK studies.
For these studies I have not used capsules but rather preferred
solution /
suspension of drug from its dispersion.
If any thing specific you would like to know, please let us know.

Torpac (wwww.torpac.com) sells mini capsules for rats in two sizes with
capacities of 25 and 80 microliter. Same diameter different lengths.
Capsugel (www.capsugel.com) sells PCcaps with a capacity of 22
microliter.
Both are relatively easy to work with as the kits come with a filling
apparatus and dosing syringe, although our experience favors the
Torpac. No
problem to use Gelucire as it is compatible with the gelatin.

Dear Marla,
you could also dissolve / disperse your drug-Gelucire mixture before
administering it orally to rats. By doing so, a gavage for oral dosing
would be sufficient, plus you=B4d probably see a faster absorption
(smaller
tmax) since the Gelucire doesn=B4t need to melt or dissolve in the GIT.

Dear Prashant,
May I know the ratio's of drug to Gelucire to be used in
solution/suspension studies for rat? Because I found some degree of
diarrhoea in formulations using gelucire as vehicle.
