I'm a first year PhD student culturing rat hepatocytes
(primary culture) using modified L15 (Leibovitz) media
supplied by Sigma in 5%CO2. I'm culturing as spheroids, so
proliferation/long term storage is not required. I'm doing
toxin testing using both traditional biochemical (enzyme)
tests, and proteomic/genomic analysis.
Q1 - L15 is not meant to be used in CO2 - does anyone know
if there are any detrimental effects from using L15 in CO2?
Q2 - Sigma have stopped manufacturing media - I'm looking
for input for a suitable alternative (DMEM already
discovered to be not as good).
Q3 - I need to use protease inhibitors to protect proteins
for proteomic analysis. How will these affect my enzyme
assays (I'm measuring LDH, GPT, GOT, GGT).
Any input would be appreciated!
