I am trying to set up a system based in fluorometric quantitation for
CYP phenotyping. I chose AHMC \0x2021 AMMC reaction (AHMC :
3-[2-(N,N-diethyl-N-methylammonium)ethyl]-7- hydroxy-4-methylcoumarin,
AMMC :
3-[2-(N,N-diethyl-N-methylammonium)ethyl]-7-methoxy-4-methylcoumarin)
for CYP2D6 activity evaluation and 390/460nm as wavelengths for
excitation/emission. I have observed that the substrate emits
fluorescence too when measuring at that wavelengths (approximately 1/6
of the product\0xEDs intensity).
Since our equipment cannot measure the excitation and emission spectra
of the compounds, could anybody tell me about them, or just if I should
measure at another wavelengths?
In case this is not possible, do you know ifthere is a better
substrate for the fluorometric evaluation of CYP2D6 activity (MAMC,
CEC)?
