Hi,Back to the Top
I am planning to measure drug concentrations in bone marrow. As I have
never worked with this matrix before I would like to know the best way
to spike bone marrow with the drug in question in order to prepare
calibration and quality control samples. How can I be sure that the
substance is uniformly distributed before I begin with the extraction
process?
I would very much appreciate any kind of help on this subject!
Thanks,
Nele

Bone marrow matrix:Back to the Top
Dissolve the drug in water; spike the matrix and then vortex for 15
seconds: sonicate for2 minutes and see if that works: the best
approach is first to use 6 replicate spikes in matrixat 3 different
concentrations (low, medium and high). Investigate accurate and
precision of assay values.
Vary sample preparation times above in accord with results observed.
