Dear all
I have a question, if a drug bound to protein plasma is decrease, then
a free drug in plasma is increase, but some of them can distribute to
tissue and may be eliminate in organ elimination. So a free drug in
plasma is not change. Although the alpha is increase. Is it correct?.
is effect of a drug always increase?

The best answer to your question is: it depends; the consequences of
the changes in the fraction unbound of a drug depend on its PK, namely
low or high extraction ratio drug, volume of distribution, and route
of administration. A good publication to read is Leslie Benet one.
