Hi everybody.
I want to know if anyone can suggest a method or a reference for
quantifying perindopril and its metabolite perindoprilate in human
plasma. Our Cmax is 12 ng/ml for a 8 mg dose and we would like to
achieve a LOQ of at least 0.5 ng/ml. We are using an SPE extraction
method to isolate the drug from plasma but are facing problems with
retention of the analyte in the C18 sorbent bed. The nature of the
metabolite is polar and I want to know what approaches can be used to
retain the drug onto the c-18 SPE bed.
What other sorbents can you suggest for this.

try to use non silica sorbent (OASIS HLB, strataX). I have experiencesBack to the Top
with SPE of ramipril, enalapril. I think perindopril will be same.
Try to use sorbent with non silica base (for example OASIS HLB orBack to the Top
strata X cartridges).
Analyt will be eluted with MeOH after purification with H3PO4 and
water. This sample preparation is suitable for next LC/MS analysis.
I have experiences with SPE extraction of enalapril (-ate),
ramipril(-ate) n I think that perindopril (-ate) will be same.
