Dear Xiao,
Using pluse-chase experiements in epithelial cells, it
has been demonstrated that the P-gp protein
trafficking from ER to membrane takes place within ~
45 mins post-translation.The precurssor P-gp is about
~120 Kda in size which undergoes N-glycosylation and
gets incorporated in the membrane as a functionally
active 170 Kda protein. During this time , the
pre-P-gp as well as the membrane encorporated P-gp
undergoes degradation by ubiquitination and
endocytosis in that order, which affects the turn-over
rate.
