Hi all
I am looking for a comprehensive review article which throws light on
pharmacokinetic and pharmacodynamic interactions in general. I have
tried pubmed but have not been able to get something satisfactory.
Any help from the group will be appreciated.

There is an excellent overview of drug interactions in the chapter by
Steve Charnick in the new book by AAPS Press (which will be out towards
the end of summer), Pharmacokinetics in Drug Development: Clinical
Study Design and Analysis. Here is the URL:
http://www.aapspharmaceutica.com/publications/press/summaries/pkdd1.asp
Also, there is a book dedicated to Drug Interactions published by
Marcel Dekker and Edited by AD Rodrigues. Here is the URL:

Any text book of Internal Medicine will have a chapter on clinicalBack to the Top
Pharmacology, Pharmacokinetics and Drug Interactions.
The one which is more extensive is Harrison's Principles and Practice of

Dear Vipul,Back to the Top
There are some articles in DDT (2000 issue).
Also please find below one more interesting article.

Another good reference on pharmacokinetic-based drug-drug interactionsBack to the Top
is the PhRMA position paper published in J Clin Pharmacol 2003 May;
43(5):443-69 as well as in Drug Metab Dispos. 2003 Jul;31(7):815-32. by
Bjornsson et. al. "The conduct of in vitro and in vivo drug-drug
interaction studies: a Pharmaceutical Research and Manufacturers of
America (PhRMA) perspective."
There is also a textbook with chapters by therapeutic class titled
"Metabolic Drug Interactions", by RH Levy, KE Thummel, WF Tragger, PD
Hansten, and M Eichelbaum (Eds). Philadelphia, Lippincott Willimas &
Wilkins, 2002.

Dear sir:Back to the Top
In general, the clinically relevant drug-drug interactions are related
with
inhibition or induction of cytochrome P-450, in these same there are
some
important review or update, such as:
Michalets EL. Update: clinically significant cytochrome p-450 drug
interactions. Pharmacotherapy. 1998;18:84-112.
Michalets EL. Clinically significant cytochrome P-450 drug interactions.
Pharmacotherapy 1999;18:84n112.

consequences
and clinical relevance of cytochrome P450 3A4 inhibition. Clin

2000;38:41-57.
Recently, in J Am Pharm Assoc appeared an excelent work:
