We used a human pbpk model for ETBE (ethyl-tert-butyl ether) inhalation
kinetics in WinNonlin some years ago, for the purpose of fitting the
metabolic parameters. It was awfully slow. We now use Berkely Madonna
which is the most user friendly software I have come across. It is fast,
inexpensive and alows fitting/optimization and Monte Carlo simulations.
The major advantages are that it is so easy to do "What if-testing",
i.e.
change different model parameters and see how this affects the time
course
(or any other output). The built-in help functions and 5-min tutorial
allow you to learn quickly to use the program. Some limitations:
hierarchical (population) modeling is not implemented and there is
limited
statistical output.
