What is the most appropriate animal species to test the
bioavailability of a drug, formulated as an enteric coated (ec)
tablet. The tablet should be developed for use in humans.
I want to test ec tablets with a diameter of approx. 5 mm and an ec
size 3 capsule (l=15 mm, diameter=5.5 mm) in a single-dose PK study.
Approx. 5 tablets/capsules will be administered under fasting
conditions. It is planned to keep the animals fasted from -10 h pre-
dose until 2 or 4 h post dose. The first idea was to use (Beagle)
dogs but I am not sure whether gastric emptying time of solid
particles of this size is too erratic.
Thus I was wondering whether the gi-transit physiology in primates
(Cyno) or mini-pigs are more human-like.
As published data are sparse, any helpful comments are welcome.
