Dear all
I am trying to find out about the IV toxicity of cremophor EL in dogs
and monkeys. Can anybody help? I know it can cause histamine release in
dogs but cannot find information on the doses at which this occurs and I
have not found any information on its effects when administered IV in
monkeys.
Any information/references would be much appreciated

Fiona, re Cremophor EL
You may be interested in the Full Prescribing Information for TAXOL
(R). It describes the hypersensitivity reaction, the pre-medication
regime used for humans, and the observation that in a specific
clinical trial the "frequency and severity of [reactions] were not
affected by the dose or schedule of TAXOL administration."

Experiences which I got from a colleague (oral communication):
using a formulation containing 10% Chremphor EL (vol/vol):
1mL preparation given iv to a Cyno (containing 10% Cremophor EL):
were well
tolerated
0.1 mL/kg preparation iv to a Beagle dog (i.e. 0.01ml/kg Cremosphor EL):
symptoms were observed close to histamine release symptoms (dog
recoverd)
Thus, I would avoid Chremophor for iv in dogs.

the paper of Gaudy JH. et al: The effect of Cremophor El in

the information on Cynos dosed with Cremophor EL is based on a single
dose.
No experience on repeated dosig is available.
