Does addition of dextran to 0.9%NaCl for replacement of the blood
withdrawn have an advantage over 0.9%NaCl only in pre-clinical
pharmacokinetic study?

Hi Xaiowei,
Well the idea is good but may influence the PK (if forms conjugate
with your compound or shows interaction) and PD of your drug. Dextran
has numerous pharmacologic effects when infused intravenously (anti-
platelet activity, anti-fibrin activity, plasma volume expansion in
hypovolemia (reason for your use), improves microcirculation by
decreasing blood viscosity and impeding erythrocyte aggregation.
Instead try replacing blood from donor rats in your PK study.

Hi Xaiowei
During PK studies we routinely replace blood from donor animals
preserved in CPDA:1 (used by human blood banks, citrate-phosphate-
dextrose-adenine) to the ratio 1:7. Blood can be stored for up to 2
weeks and as well as being practical (can be prepared up front of
study) it saves on donor animals as the blood can be used for several
studies. We have used this for virtually rodent PK studies without
problem.
