what is the rule of tumb for dosing rat compared to human. I know that
it should be between 5 to 10 times of human dose but I need the
reference. Any reply with the references would be appriciated.

It depends on the therapeutic use of your drug. In cancer research,
you can use the animal dosage calculations proposed by the FDA :

The conversion factors are: Human dose (mg/m2) = rat (mg/kg) x 6
Human dose (mg/kg) = rat (mg/kg) x 0.15
