Dear Member,
I would like to know whether it is possible for a drug substance to get
precipitated in vivo due to the common ion effect in stomach due to the
hydochloric acid present . Further for drug showing common ion effect
invitro in 0.1HCl and having a very specific pH dependent solubility
,what
would be impact on the absorption of drug.

Dear Manoj:
Usually salts of weak base tend to get precipitated in stomach due to
common ion effect. However, effect of such precipitation on
absorption is depend upon "solubility product" of the salt in the
aqueous environment, redissolution of precipitate in the upper part
of GI (especially in presence of bile micelles), particle size of the
precipitated drug and intrinsic solubility of the free base. If the
drug perse has good oral bioavailability, common ion effect may give
delay in Cmax but no change in AUC. You need to consider effect of
food on BA of drug too in such cases.
