Dear all,
Is it feasible/ethical to administer a bolus intravenous dose to
cannulated rats (femoral/jugular) where the drug is dissolved in
saline at a pH of 3.0-4.0?

You have to be very careful when you administer a bolus intravenous dose to
cannulated rats where the drug is dissolved in saline at a pH of 3.0-4.0.
The low pH itself might not be too bad. However, the solubility of the
compound you administered could be a big issue. The pH of plasma is about
7, upon bolus iv dosing, the compounds might precipitate and hence cause
serious effects.
My suggestions:
1) Dilute your dosing solution with plasma and see if there is
precipitation. If you cannot see precipitation, bolus iv might be OK. The
protein binding might enhance the apparent solubility.
2) Use iv infusion in stead of bolus iv.
