Dear all,
I want to conduct a therapeutic study in monkeys by administrating the
test substance orally once a day over 1 year. I heard that
administration of tablets or even a solution (by gavage) is not
recommended in monkeys. An alternative is to incorporate the test
substance in a cookie. I'm not very convinced by this route of
administration as it is not representative for the route of
administration in man and also, it is not the better way to determine
the PK parameters (other contents of the cookie, homogeneity, size - )
Does anyone have any experience with oral administration in monkeys?

There is no problem dosing monkeys orally with tablets or by gavage,
even for one year (or longer).

It is common practice in toxicology studies to routinely administer
solutions or suspensions by gavage for long periods (intragastric via a
rubber cannulae) monkeys need to be acclimated to the procedure for ca
one week prior to initiation in a way to exclude the animals that does
not respond well to the procedure itself. Incorporation in food is
always a possibility with all the variability that you can imagine.

We used to solve the drug into the drinking water for
monkeys and measured how much water left in the bottle
to calculate how much they took per oral route.

Dear Samia,
the best for oral dosing in monkeys is probably a gavage formulation.
People working with monkeys have no problem dosing PO.
Why don't you try an oral suspension ?

gavage administration of solutions/suspensions to marmosets [ where a
treat of
something sweet afterwards makes the medicine go down]
to
Capsule administration to baboons and cynomolgus monkeys [ by well
trained staff]
to
watching long term work [study duration >5 years !] at a CRO where
toxicity of
oral contraceptives involved single daily administration of a tablet
and dosing
involved opening the cage door, the chip came out and took her own
tablet, got
her reward and went back into the cage.
