Dear All:
I'm searching papers about PAMPA-BBB, I would like to know the
composition of the lipid that mimics BBB and who is the provider. I've just
bought the PAMPA Bundle. Is there any way to modify the lipid in the PAMPA
Bundle (GIT) in order to mimic BBB? Thanks a lot!
Best Regards,

Dear Ixchel
There has been some work done on Pampa for BBB. In the original PAMPA
assay, lipids were chosen to reflect intestinal lipids, however, in this
modified version, porcine polar brain lipids have been utilized to better
reflect the BBB. These lipids are available from Avanti Polar Lipids. A
great reference for this is:
Eur J Med Chem. 2003 Mar;38(3):223-32.High throughput artificial membrane
permeability assay for blood-brain barrier.
Di L, Kerns EH, Fan K, McConnell OJ, Carter GT.
This reference details the lipids and their percentage composition.
However, it is important to remember that this assay is only useful to
predict the transport of compounds which pass the BBB via passive
diffusion, and the effect of efflux transporters, active uptake
transporters, are ignored.
Hope this is of use for your work.

Dear Ixchel,
you may want to request via pION's website (http://www.pion-inc.com/) a
poster that describes a BBB permeability model based on PAMPA measurements,
incorporating a learning set of nearly 300 log PS values. It states on the
site that a full paper will be submitted soon.
Kind regards,
