Dear Group,
We run a standard in vitro discovery screening (10 fold dilution) to
determine whether chemicals are mechanism-based inhibitors of CYP
isozymes. Sometimes, we see time-dependent inactivation but NOT
CONCENTRATION dependent. Has anyone ever seen this phenomenom and if so
it seems that this type of chemicals does not meet the criteria to be
defined as a mechanism-based inhibitor, but what could be a possible
cause for this type of inactivation?
