Dear
I am pursuing Ph.D in pharmacy in jadavpur university. i am working
on glimepiride. I have faced lots of problems on dissolution of
glimepiride.I have tried the following as dissolution medium,
1. Phosphate buffer Ph 7.8 +1% SDS
2. phosphate buffer Ph 6.8 +1% SDS
3. phosphate buffer Ph 5.5 + 1%SDS
4. phosphate buffer Ph 7.8 +1%Brij
5. Phosphate buffer Ph 6.8 + 1% brij
6. phosphate buffer Ph 5.5 +1% Brij
But results are not satisfactory. please help me.

Dear
before selecting the particular dissolution medium need to be done
apparent solubility studies at 37*c.
In USP also given the selection of dissolution medium or
www.usfda.gov/cder/guidance.

Glimepiride has very low aq. solubility. But around 70-80% release is
achieved in pH 6.8 Phosphate buffer and nearly about 90% in pH 7.2
phosphate buffer. Even most of brand and generics available also
shows similar dissolution profile and are proven to be bioequivalent.
