Dear all
I am a novice on building ppk model. I have found that some paper
said the pbpk model could be used as the structural model in the
NONMEM model. But i can't find any material on how to do it. Could
anyone give me some information? Thanks!

Dear,
In order to estimate population variability in a PBPK model you need two
skillsets: (i) setting up a PBPK model using differential equations and
(ii) estimation of pop-PK parameters using Nonmem. Neither of those
skills are easy to learn properly. I imagine the combination is even
harder (I have never done that myself). If you have never done popPK, I
would suggest following a course in that area. An alternative is perhaps
doing an internship at a group like that of prof. Rowland, who is active
in research in that area. Anyway, you should brace yourself for quite
some investment in time/money.

Hi,
as far I know there is very few published attempts to combine PBPK
and popPK. I've seen a PhD thesis written by Jonsson from Uppsala
University (you can download it on their website) and the papers of
F.Y. Bois and colleagues (see Medline).
I hope this could help you, but I agree with Jeroen : be prepared to
a long long work...

You may want to check out this recent paper by Gueorguieva, Aarons
and Rowland in JPP (2006,33:5,571-94). Winbugs (and PKBugs) are
available on line free of charge and they provide considerable
modeling power and flexibility.
