Dear all,
i want to conduct in vitro metabolic study of atorvastatin using
human liver microsome preparation.
1. Can i use atorvastatin calcium instead its pure acid form?
2. Can we dilute the liver microsome preparation with potassium
phosphate buffer [pH 7.4] and store it for a long period?
3. What are all precautions and care to take while performing these
type of studies?

Salt form can be used.
The microsome preprations are frozen for long term storage and
periodically thawed for experiments.
During metabolism experiments, microsomes are added to reaction
mixture containing phosphate buffer 7.4 and NADP regenerating system.
You can keep the microsomes in 7.4 buffer at 37oC with the NADP
regenerating system during the experiment.
It is not recommended for long term storage.

You can very well use atorvastatiin Calcium in place of atorvastatin
acid
provided you should also account for the calcium content while preparing
your stock solution.
Secondly, it is not advisable to store diluted stocks of microsomes
in the
assay buffer.
FYI lactone form of statins are more easily prone to metabolism than
acid
forms, therefore confirm about atorvastatin calcium, whether it
exists in
lactone or acid form.
I would appreciate experts views on this aspect.
