Dear all,
I would like to have explanation on my doubt. In the process of HPLC
method development for determination of insulin in rat plasma, I have
done extraction with 3 ml of dichloromethane and reconsitituted with
0.1 N HCl, then evaporated dichloromethane. I have got recovery about
30%. When the supernatant of organic solvent evaporate to app. 200
ul, then reconsititue and evaporate remain amount of dichloromethane,
recovery was improved to 60%.
I need reason for it. As i expected that "it might be due to low
transfer rate of aqueous phase into organic phase in the presence of
higher amount of organic solvent". Is it correct?
I will appreciate if members can provide information on it. Any
reference will be very useful.
