This is a recent publication which, I believe, will answer your
second question. While it focuses on anticancer agents in humans, it
includes an informative introductory review of general interest.

dear santhosh
radiolabeling is mandatory studies for pk studies to submit the FDA
submission. This radio labeling can tell the exact amount of drug
distributed in every organ, need not drug to be extracted.
You can find lot of references in www.science direct.com

Hi Santosh,
Radio-labeled compounds are widely used to investigate absorption,
distribution, metabolism and elimination (ADME) of drugs during
discovery and development. Accelerated Mass Spectrometry (AMS) is
currently being used to measures tracers such as 14C. With such ultra-
sensitive techniques, it is possible to lower the radioactive dose
and conduct studies in human beings.
