We use the orbital sinus extraction of blood in rats and mice as a
common
procedure. In rat we got 6 points ( 3 for each eye 50-100 uL). With this
method we had obtained normally a very good Pk profiles. But now we
have
problems with animal ethic welfare committee and they want to eliminate
this procedure due that is consider very cruelty. Do you have the same
problems? What do you think is ethic or no? Alternatives for TK studies
o PK studies?

Dear Benjamin,
I think the most ethical method would be canulation of jugular vein
day before the experiment.
If proper anesthesia and analgesia are used for the surgery, there is
a minimal post-surgical pain expected. The sampling is pain-free and
much less stressful then any invasive method.

It has been my experience that since about 5 years ago Institutional
Animal care and Use Committees have generally limited orbital bleeds to
only anesthetized animals and one sample per eye with a final terminal
sample for a total of 3 per animal unless the sampling times were days
or weeks apart.
