Hi All,
We did a rat IV infusion study. The results were interesting and
puzzling. We set up the infusion for 15 min. The vehicle is 60% PG
and 3 % Dimethylacetamide in saline. Blood samples were taken at 5,
10, and 15 min for the infusion phase. The plasma concentration was
highest at 10 min instead of 15 min when the pump was stopped. The
concentration at 15 min was about 50% less than that at 10 min. We
double checked infusion pumps. Pumps seem accurate. The plasma
concentration after 15 min seem good (like regular IV bolus). Did any
body encountered the same situation before? How can I interpretate
the data?
Any suggestions will be greatly appreciated.

Dear Jason,
Have you taken the concn time profile for some more period. that
would have given you a more detailed picture.
I cannot conclude basing on the little info you have given. Does the
drug has first pass effect. or May be there is a rapid uptake by the
followed by equilibration with the tissue compartments.
There are many thought domains before a possible answer can be found.
