Does anyone know about or have experience on PK variation of
metformin with slow-release formulation according to the dosing time,
like morning versus evening administration? Beside to reducing GI
adverse effect, evening dosing is known to increase systemic exposure
of metformin compared to morning. And it is believed that metformin
would be more beneficial in the evening because gluconeogenesis
increase during the night. I would appreciate any references, reading
materials, or any experiences you have had about these.

No direct data on this. However
Improved nocturnal absorption likely to be due to reduced competition
for OCT-1 transport ie relative increased oral availability.
PK in hepatocytes very different to plasma with much less variability
and unlikely to be clinically significant, ie nocturnal dose unlikely to
have gluconeogenesis effect over daytime exposure, but would be an
interesting experiment.
