Dear PK Group Members,
I have 2 questions regarding N-acetyl transferase enzyme (NAT 1 and
NAT2).
Do these enzymes have circadian rhythm?
Are these enzymes expressed only in liver?
Thank you in advance.

Dear Raju,
Some N-acetyltransferase enzymes are also present in other organs,
e.g. the
small intestine. Have a look at www.interspeciesinfo.nl

NAT1 is expressed in most tissues of the body - including liver,
urinary bladder, colon, mammary gland, lung, kidney, small intestine
and pineal gland -- whereas NAT2 is mainly expressed in liver and
intestine. However, most (but not all) of the tissues that express
NAT1 also appear to express low levels of NAT2, at least at the level
of mRNA (Rychter et al., 1999). N-Acetyltransferases are cytosolic
enzymes found in liver and many other tissues of most mammalian
species, with the notable exception of the dog and fox, which are
unable to acetylate xenobiotics.

Re NAT Expression
Dear Raju,
NAT1 also express in lymphocyte. Therefore lymphocyte lysate can be
used as the source for NAT1
phenotyping. See detail in " Kukongviriyapan V et al. Polymorphism
of NAT1 genotype and phenotype in a Thai population. Eur J Clin
Pharmacol 2003; 59:277-281 "
