I would like to know any theories / methods that be used to predict
the optimum concentration of a buffer for mobile phase preparation in
Chromatographic seperation.

Without knowing your analyte and instrumentation (e.g. UV-HPLC vs. LC/
MS) I
hesitate giving broad-spectrum advice. But, below are some places I
go when
I need to refresh on "buffer-ology."

Experts from the
chromatography field, end-users, and vendors all contribute to the
discussions on the site.
http://www.zirchrom.com/pass.asp has a buffer wizard.
http://www.mac-mod.com/pdf/TR-BufferedMP.pdf is a Mac-Mod report on
preparing mobile phase buffers for reversed phase chromatography.
