Pka of the saquinavir is 7.1

Saquinavir has two base pKas one about 2 and one about 7. This
produces a
complex solubility vs pH curve, and can influence permeability as
well. The
pKa at 7 results in decreasing solubility (and increasing
permeability) in
the small intestine as the drug is less and less ionized going from
duodenum
to ileum.
There is a third pKa (acid, about 11) that has no effect at
physiological
pH.

Dear WALT,
Truly agreed regarding the PH dependent soubility and permeability of
Saquinavir.
We have observed the same with Quetiapine.
But can we have this same phenomenan for all drugs? Or depends upon
whether drug is acidic or basic in nature?
I suppose saquinavir(as a drug not formulaiton) is basic in nature.
Please clarify.

It depends on the number of protons available for dissociation or
association (Lowry-Bronsted theory).
